Wilson will support agents in Scotland and Northern Ireland while Eastwood will help agents in the north of England.
Eastwood has a strong background in retail and business development, having held roles at Thomas Cook, Hays Travel, Barrhead Travel, Cruise118 and, most recently, Red Sea Holidays.
The line said Wilson "brings a wealth of cruise industry experience and a deep commitment to creating unforgettable journeys".
She has been tasked with building new relationships with the Northern Irish trade as well as being responsible for managing the Scottish Passenger Agents’ Association (SPAA) relationship in Scotland.
Both Eastwood and Wilson will report into Lynsey Roberts, business manager for partnership and regional development.
P&O Cruises said it hopes the expanded team can "enhance its regional presence and strengthen relationships with the trade".

Both Wilson and Eastwood will kick off their roles hosting agents on ship visits on the line’s Aurora vessel in Liverpool and Greenock, followed by visits to their accounts’ head offices in their respective regional areas.
