The daily link from Dublin to the Connecticut city will be summer-only, restarting on 26 March and operating until the end of October. One-way fares will start from £229.
Hartford, located between Boston and New York, is known as the insurance industry capital of the world, but is also home to the Mark Twain House and Museum and is near Yale University.
Aer Lingus began flying to Hartford in September 2016 but suspended the service in 2020. The destination will be the airline’s 16th transatlantic route.
Lynne Embleton, Aer Lingus chief executive, said: “The restart of the daily flight into Bradley International airport is the only direct service to Europe, reflecting Aer Lingus’s ability to connect UK and European customers with North America.
"Aer Lingus is forging ahead with its North American expansion plans, growing the number of transatlantic routes from its Dublin hub."
The Dublin-Hartford service will be operated by a single-aisle Airbus A321neo offering business and economy class.
