Applications for the seven-night trip, which departs on 16 October, are open until 2 September. The successful applications will fly direct from Manchester onboard one of Tui's Boeing 787 Dreamliner aircraft.
Tui said the trip, for which it has partnered with Universal Orlando Resort and Visit Orlando, has been designed "to give agents first-hand experience of one of the world's most popular holiday destinations, helping them build their knowledge and confidence when selling Orlando holidays to customers".
Agents will spend three nights exploring International Drive, which Tui said was one of its "top-booked areas" outside the major theme park resort locations in Orlando, experiencing its attractions, hotels, restaurants and entertainment.
The group will then spend four nights at Universal Orlando Resort, experiencing a range of hotels, attractions and guest experiences. The journey will be another feature of the trip – the outbound experience at Manchester airport, the flight and arrival into Melbourne Orlando International airport.
Andrew Isherwood, Tui UK and Ireland's Head of Trade Relations, said: "Orlando continues to be a favourite destination for UK holidaymakers, and this trip will showcase everything from the energy of International Drive to the world-class hotels and attractions at Universal Orlando Resort.
"Experiencing it all first-hand will equip agents with valuable insights and stories they can share with customers, helping bring holidays to life and inspire future bookings.”
Applications are open via this online form to independent agents over the age of 21 working in sales roles. Applicants must not have visited Orlando in the past five years, and will need a valid passport, Esta and travel insurance.
