Three-times-weekly Heathrow-Larnaca flights will get under way on 5 November, flying on Tuesdays, Fridays and Sundays.
The route will be operated year-round using a 144-seat Airbus A319 aircraft, with London becoming the airline’s 11th international destination.
Earlier this month, the carrier inked a deal with Surrey-based Flight Directors to serve as GSA and to allow it to set up a UK office.
The airline said the aim of enlisting Flight Directors’ assistance was to boost brand awareness in the UK.
It is currently finalising plans for travel agency distribution, with Flight Directors responsible for UK sales development, reservations, ticketing and additional customer service assistance.
Cyprus Airways also hopes to leverage "strong, established ties" with specialist tour operators.
Chief executive George Mavrocostas said: “With Flight Directors as our new partner, we will significantly strengthen our sales team and drive sales within the travel trade.”
Paul Argyle, Flight Directors managing director, added: “This is a real, positive sign of the market for air travel returning. The new service is starting before the summer ends and will continue throughout the winter, satisfying part of the pent-up demand for sunshine holidays.
"The support of the UK travel trade is important to the success of the new route and we are committed to developing strong ties with all travel organisations interested in booking passengers to Cyprus."
